Crude adenovirus solution contains recombinant adenovirus ready to amplify for expressing the protein of interset. Using 100µl solution, infect the cells culturing in 100 mm dish and observe propagation.
The solution contains approximately 1.0 x 109 pfu/ml adenoviruses ready to amplify and purified with the Adeno-purification Kits (Ad1001, AD1002 & AD1003). The virus is replication-defective based on human type-5 adenovirus. It can only be amplified in the cells expressing adenovirus E1 proteins such as HEK293 cells.
To amplify, infect HEK293 cells grown in 100mm with 100µl crude adneovirus solution. It will take 2-3 days to make all the cells dead and floating, and collect cells and medium together. Briefly centrifuge and use 100µl supernatant for further amplification or store at 4°C. If purified adenovirus, store at -20°C.
